App Note: High-Throughput Single-Cell Genome Profiling

Genetic heterogeneity in tumor tissues is a key factor underlying drug resistance and metastatic potential. While single-cell methods provide a valuable tool to dissect this heterogeneity, to date, these methods have been limited in their throughput. The new Shasta™ Single Cell System, combined with PicoPLEX® whole genome amplification (WGA) technology, overcomes this challenge. This app note demonstrates how the complementary Shasta and PicoPLEX technologies enable automated generation of high-throughput single-cell DNA-seq libraries for sensitive CNV and SNV profiling to identify tumor-driving mutations.
